Versatile Automation System

The customer presented the Project Management team with the need to set up a versatile automation system that could accommodate the multiple assays run in the lab. Specifically, the customer needed to automate the screening of their core compound collection (150-300 compounds) for binding to targets within a protein family.

A Biomek® Assay Workstation was designed and installed to enable the automation of four assays. The assays include a Nuclear Hormone Receptor assay, a Protease/Phosphodiesterase Assay, a Tyrosine Kinase assay and a Serine/Tyrosine Kinase assay. All four assays have been designed to be mix-and-read homogeneous assays so the key deliverable for the system was provision for all reagent transfers and incubation. Data collected from each of the assay sets will be used to enable medicinal chemists to design modifications to the compound structures that will increase their binding affinity to the targets.

This customer chose the dual-bridge Multi-Multi Biomek FX linked to a Cytomat* Microplate Hotel for additional labware storage. The Barcode Reader along the Cytomat ALP allowed for sample and labware tracking throughout the assay. The Biomek FX Multi-channel Selective tip pipetting capability and SAMI® EX were also incorporated into this Assay Workstation. Given the homogeneous nature of the automated assays, the customer found that the ability to access variable rows and columns using the selective tip option on the Multi-Channel head was faster and less expensive than the installation of a Span-8 pod. This solution satisfied the customer’s high demand for precision and accuracy. SAMI EX software enabled the customer to schedule the run time and maximize throughput for their assays.

The system has met the customer’s requirements and continues to perform satisfactorily.
