BiomekŪ Assay Workstation Integrated with Two Readers

The customer, a drug safety group within a major Pharmaceutical Company, presented the Project Management team with a need to automate their processes involving the assessment of toxicology for new drug candidates. Their methods included creating an irregular dilution pattern to determine IC50 values using three different assays to determine toxicology.

To satisfy their needs, the project management team designed a Biomek FX-based Assay Workstation with a Cytomat* MPH and a Cytomat 2C-450 shaking incubator. Both Cytomats were linked to the deck of the FX via a Cytomat ALP with a Bar Code Reader. To manage the use of fluorescent and luminescent cell-based assays to determine drug candidate toxicity, the team provided a custom integration of Labsystems Fluoroskan and Lumiskan Ascent readers. The readers were linked to the FX deck via BRT, robotic transport and the Cytomat ALP. This custom integration of the two Labsystems readers via BRT was novel for the project management team, but it allowed the customer to run their three cell-based assays on the same platform at once. The team was able to add value for the customer by setting up the scheduler to interleave the three cell-based assays and optimizing the overall throughput of the system. The final component of the system was the Selective tip software and modified receivers that enabled the customer to generate the dilution pattern (right to left on the plate) for IC50 determination. The configuration of the Assay Workstation is shown below (Figure A):

The customer chose the Beckman Coulter solution in a competitive sales situation based on the installed base of reliable instruments on site at this Pharmaceutical company. The ease of use for both SAMIŪ and Biomek FX software was also a contributing factor.

The customer is extremely happy with the design and performance of this system. An alternative multimode reader could have worked, but the customer’s protocols were already validated on the two Labsystems readers, they did want to take the time to spend validating a new reader before the system purchase.
