BiomekŪ FX Specification for Integration of Serial Dilution Step

Overview

This Custom Serial Dilution Step has been written for the BiomekŪ FX to utilize a 96- or 384-Channel head for serial dilution across a single plate. The Multichannel head will be loaded with one column of tips along the left side (Column 1) of the head, or with one row of tips along the backside (Row 1) of the head. The rest of the head will be loaded with shorter tips*. The longer tips will be utilized for the serial dilution process.

The warranty for this integration is one year from shipment. This warranty applies for the integration to the version of software originally integrated. Any changes to system components that cause the integration to no longer function properly are not covered under this warranty.

The warranty applies to the system and system software versions on which the original integration was installed. Although Beckman Coulter's goal is for the integration to be forward compatible, this cannot be guaranteed for custom integrations. Any upgrades to the system that cause the integration to no longer function properly are not covered under the warranty for this integration.

Functional Description

This integration allows the customer to perform serial dilutions across a single 96- or 384-Well plate on a Biomek FX utilizing a Multichannel head. The serial dilution can be performed across the rows or columns of a plate. If using a 96-channel head, AP96 200 tips must be loaded on row 1 or column A*. All other tips must be AP96 20 tips. With a 384-channel head, AP384 30 tips must be loaded on row 1 or column A. All other tips must be tips that have been cut off. In order to load tips on the Biomek FX, a full tip box must be used*.

The following variables may be set for the serial dilution:

  • Pod to run the serial dilution step
  • Tip type with option of unloading or keeping tips on after transfer
  • Wash tip option with number of cycles per wash; washing after transferring diluent; washing after transferring source compound from master plate, and washing when finished with serial dilution step.
  • Plate position to perform dilutions
  • Dilution direction (left to right or top to bottom)
  • Start and end (row or column) of dilution
  • Volume and liquid type selection
  • Technique selection for both the dilution, diluent, and source compound (similar to Biomek FX aspirate and dispense techniques)
  • Discard excess volume and deck position of dilution
  • Abilities to add diluent before transfer, and to the left-most column
  • Diluent position on Biomek FX deck
  • Liquid type of diluent
  • Labware type in which holds diluent**
  • Dilution ratio
  • Master plate position
  • Labware of master plate
  • Overriding auto-selected technique can also set other variables such as tip touch, blowout, mix count, speed and volume.

This integration will support serial dilutions across 96- or 384-well plate types supported by the Biomek FX software.

Device Control

All tasks are controlled through the Biomek FX software. The Multichannel pod serial dilution step can run as a single or multiple steps within a Biomek FX method. No additional hardware or software is required*.

Data Collection

There are no data files generated from the process, other than a Biomek FX standard log file showing general tasks performed during the method operation.

* The modified tip arrangements described within these specifications are not necessary if the user purchased the custom Biomek FX Modified Receivers that allow the loading and unloading of single row or column of tips from a standard Biomek FX tip box.

** If aspirating from a reservoir, liquid level tracking may not be accurate due to the fact that the Biomek FX software operates as if it is aspirating or dispensing a full array of tips on the multichannel head. Therefore, if aspirating, the tips may lower faster in the liquid; if dispensing, the tips will rise faster than the liquid.
