Biomek® 2000 Specification for Integration of Hit Picking Wizard Software

Overview

A Hit Picking Wizard software utility will be developed for the Biomek 2000 Laboratory Automation Workstation. The wizard will allow the operator to specify how hits in row/column format are transferred to a destination plate. A worklist input file will specify the hit wells of the source plates. The Hit Picking Wizard will generate a BioWorks™ method based on the worklist input file and operator specified information. The operator can then run this method. A Destination Plate Map file and an Event Log file will be generated at run time. The Destination Plate Map file will list the well location and source plate ID for each well on the destination plate. The Event Log file will contain a list of bar codes of the source and destination plates processed by the system. The Hit Picking Wizard software for the Biomek 2000 will be created by Beckman Coulter.

The wizard will consist of a series of screens in which user input will be taken. Within the wizard, the operator will select between Reformatting or Hit Picking. For Reformatting, the source pattern and destination pattern are defined for the transfer. For Hit Picking, a worklist input file will be used, specifying a list of wells in bar coded source plates to be transferred to destination plate(s). After collecting input, the wizard will create a Biomek 2000 method. This method will contain the steps necessary to move the source and destination plates from the stacker carousel to the deck of the Biomek 2000 (if selected), transfer the hits from the source to the destination (or perform reformatting), and return the plates to the stacker. An event log will be kept and a file listing the contents of the destination plates will be produced. In the case were no stacker carousels are used, all plates and tips will start on the deck.

The warranty for this Custom Integration is one year from shipment. This includes Beckman Coulter- supplied hardware and software. This warranty applies for the integration to the version of third-party hardware, firmware, and software originally integrated. Any changes to the third-party component that cause the integration to no longer function properly are not covered under this warranty.

The warranty applies to the system and system software versions on which the original integration was installed. Although our goal is for this software to be forward compatible, this can not be guaranteed for custom integrations. Any upgrades to the system that cause the integration to no longer function properly are not covered under the warranty for this integration.

Functional Description

The operator will provide a worklist file for the run. The worklist file will contain the plate number, plate barcode, hit well location in row/column format, volume to be transferred (optional), and well content ID (optional). This file will be specified in the Hit Picking Wizard interface. The operator will place on the deck, or load in the Stacker Carousel the plates specified in the worklist file in the exact order that they are in within the worklist file.

The operator will start the Hit Picking Wizard and specify the worklist file to be used for the run. The following options will be supported:

  • Source and Destination Plates - The operator will specify the type of input source plate and the type of output destination plate. Only one type of source plate and one type of destination plate can be specified. Only unlidded plates can be used. The source plate type and destination plate type can be different, however. Only 96 and 384 well plate types are supported. Special plates types not included in the Biomek 2000 software library are not supported.
  • Volume for Transfer - The operator will specify the transfer of a fixed volume for each well, or to use the volume specified in the worklist file.
  • Reformatting - Instead of providing a worklist of hits, the wizard will allow the operator to specify a source plate pattern to be used for each source to create a list of wells to be transferred. A pattern will also be selected, or specified, for the destination plate(s).
  • Deck Layout - The deck layout will be specified in the Hit Picking Wizard. The wizard will write a BioWorks method to perform the hit picking, therefore, it must know the deck layout.
  • Support for a Full Plate of Hits - The maximum hits per plate will be equal to the maximum wells in the plate. A full plate of hits in a source plate can be transferred if desired.
  • Support for Stacker Carousels - The option to use only the deck of the Biomek 2000, or the Stacker Carousel(s) can be selected. Stacked plates are not supported. The stacks to use for the source and destination plates is selectable. P20, or P200 tips can also be used from the Stacker Carousel. P1000 tips can only be used on the deck of the Biomek 2000. None, one, or two Stacker Carousels can be used.
  • Support for Bar Code Verification - When not using Stacker Carousel(s) or for plates starting on deck, the bar codes of the plates can be verified with a handheld bar code scanner. The software will walk the operator through scanning in the source and destination plates in this case. When using Stacker Carousel, the bar codes are verified during the run with the option to stop if the bar code in the worklist does not match the bar code of the plate expected. The bar codes can also be logged to a file as they are read. Bar code verification or reading can be turned on or off.
  • Serial Dilution NOT Supported in Wizard - Serial Dilution can be performed in a separate BioWorks method. It is not supported as an option within the Hit Picking Wizard software. Full BioWorks software functionality remains available for various pipetting tasks.

Note: Reloading of the Stacker Carousel(s) during the middle of a BioWorks run cannot be utilized at this time. Only one Stacker Carousel load at the beginning of the method run, with included empty stacks for returning plates (or tip boxes), can be supported. If more source or destination plates are used that can fit in the Stacker Carousel(s), then an error message will be displayed requesting that fewer labware (a shorter run) be specified. Please edit the Worklist File to reduce the number of plates or tip boxes that are retrieved from the Stacker Carousel. This does not apply for a system without Stacker Carousels.

This integration includes the Hit Picking Wizard software only as a custom option for the Biomek 2000 liquid handling system.

Data Collection

The Hit Picking Wizard will generate two data files. These files will be transferred prior to the run, or during the run as necessary. These files will be comma separated text files that can be used for importing into other utilities, such as Microsoft* Excel, for analysis. A Destination Plate Map file, and an Event Log file will be generated.

  • Destination Plate Map File - The destination plate map file will contain the destination plate information after the transfer has taken place. Each line in the file will contain the Plate number of the destination plate, the bar code ID (if available), the well location (Row/Column) of the destination plate, the source plate number, the source plate bar code ID (if available), the well location (Row/Column) of the source plate transferred, the volume transferred, and well content ID (if available).
  • Event Log File - This will be a list of the events or tasks that the Biomek 2000 performs. It will include bar codes read during the run. It will contain a top to bottom list of events in the order that they were performed during the run.

Additional Specifications

Microplates The Biomek 2000 can use a variety of manufactured 96 and 384 well plates. Only types supported by the BioWorks software are supported by the Hit Picking Wizard. Tubes and Tube Racks are not supported.
Bar Code Reading A Handheld Bar Code Reader, supported by the Hit Picking Wizard, is optional. Bar Code Readers mounted on the Stacker Carousels, supported by the Hit Picking Wizard are also optional.
Hardware This specification only covers the Hit Picking Wizard software. No special Biomek 2000 hardware is included or required. The Hit Picking Wizard uses standard Biomek 2000 hardware, and standard Stacker Carousel configurations.
