Horizontal Loading of Carousel and CO2 Incubator: SAMI Tips and Tricks

Question:
Is it possible to schedule a method, so that the plates in the CO2 incubator are stored horizontally rather than vertically?

Answer (by Christoph Krüll, Ph.D, Beckman Coulter):
This is possible. The following description assumes that a SAGIAN™ CO2 Incubator and SAMI version 3.0 or higher is used. A Deck Carousel or a Heraeus Cytomat can be configured in the same way; only the slot numbering looks different.

Fig.2 Modify Property configuration. Double click on the Modify Prop' icon in the SAMI method view (Fig.1). Configure the node to Action 'Set', define the name of a new Property (here 'Enzyme') and assign the Default Value 'A'. For the 2nd assay plate (AssayB) set the Default Value to 'B'.

Open SAMI software and select the menu entry Palettes/Station Builder. Then click the button NEW in the Logical Stations field, enter a name for this station, e.g. CO2Horizontal, and confirm with OK.


Now we want to configure the station in the Logical Station Definition section. Under Parts it should show Empty Set.

Go down to the Station field and select CO2Incubator1. Empty Set will now change to CO2Incubator1 1-180 and all 180 slots will show in blue on the right-hand side under Slots. Select only slot No. 1 at this time by clicking on 1 in the Slots field, then hold down the CTRL key and select slots 19, 37, and 55.


We just defined a station with four horizontal positions. SAMI will use it in order of the numbers, i.e. 1, 19, 37, 55. To define the second horizontal layer, click on the New Part button and again select the 'CO2Incubator1' in the Stations field. Then select slot No. 2, hold the CTRL key and select slots 20, 38, 56. Continue for all horizontal shelves you want to use, maybe all 18. After three rows or shelves it should look like this.

It is very important to define one horizontal line after the other. SAMI will use the slots in the order as defined in the Parts field. Here, all available slots in the pattern defined between slot 1 and 55 (i.e. 1, 19, 37, 55) will be used first. Only then SAMI will go for the next part, the pattern between slots 2 and 56 (2, 20, 38, 56). Click OK when the definition is completed.

Now all you have to do is open your SAMI method, double click the source icon and reconfigure it from CO2Incubator1 to CO2Horizontal in the Home Stations field.

If this definition is used more frequently, just change the source icon definition within the SAMI palette by double clicking onto it. You can also use the Palette Editor to create a new icon for this source.
