iTox 3 System: Standardized and Automated Rat Immunotoxicology Using the BiomekŪ NXP Span-8 and the Cytomics FC 500 MPL

Beckman Coulter recently released two flow cytometry three-color combinations (combos) for rat lymphocyte immunophenotyping in preclinical immunotoxicologic applications:

  • IOTest_ Anti-Rat CD3-FITC / CD4-PC7 / CD8-APC (PN A32909)
  • IOTest Anti-Rat CD3-FITC / CD45RA-PC7 / CD161a-APC (PN A32910)

These new “iTox 3” cellular analysis combos have been developed for Beckman Coulter's Cytomics FC 500 series flow cytometer with dual-laser configuration and multiplatform loader (MPL) option.

Standardized, easy to handle and manufactured under cGMP conditions, the iTox 3 combos have several advantages: they provide more information with less sample than one- or two-color offerings and the number of analyzed parameters for the same volume of sample can be further augmented with a choice of drop-in reagents (e.g. Anti-Rat CD45-PE, PN A36700). In addition, the adapted no-wash red blood cell lysing procedure streamlines sample preparation compared to implementing manually.

For high-volume applications, the sample preparation and lysing process were automated on Beckman Coulter's Biomek NXP Span-8 instruments. This automated workstation puts every aspect of liquid handling – including pipetting, diluting, dispensing and combining samples with reagents – into a single, small footprint
device. Coupled with an intuitive software interface, the methodology relies on easy and flexible procedures, whatever the samples to be handled.
