Comprehensive Mapping of Protein Expression Profiles in Activated and Non-Activated Peripheral Blood Mononuclear Cells

The ProteomeLab™ PF 2D platform enabled the comprehensive analysis of cellular proteins in SEB/CD28 activated and control PBMC cultures at 6 hours post-activation while easily homing in on the proteins relevant to the activated culture. The preliminary identification of protein(s) playing an important role in activation of an immune response was accomplished in a single day using the completely automated PF 2D platform. The profiles of proteins differentially expressed between control and activated cultures determined that activation was associated with a loss or down-regulation of acidic proteins while increasing the expression of selected basic proteins. Similar studies can be done by sampling the control and activated cultures over a period of time to gain an understanding of the ongoing processes of activation. In addition, supernatants of the cultures can also be analyzed to identify differences in secreted proteins between control and activated cultures.

In our experience, the PF 2D platform enabled the automated, rapid, and effortless mapping of complex protein expression patterns, while serving as a perfect complement to the currently available techniques of ELISAs, Western blots, Edman sequencing, capillary electrophoresis, and mass spectrometry to characterize the proteins of interest. These characteristics make the PF 2D platform an ideal tool for use in a wide range of applications from drug discovery and development, clinical research and trials, to the basic research and identification of novel proteins.
