Poster Abstract

Automated fluorescent sequencing using ABI PRISM* BigDye* Terminator Chemistry is a common method for high-throughput DNA sequence determination. Common purification methods are based on gel filtration and/or precipitation by ethanol. Promega has developed a series of ‘silica-paramagnetic’ particles (MagneSil) for the purification of nucleic acids from reaction components in the BigDye Terminator DNA Sequencing reactions prior to automated sequence analysis. This removal prior to analysis enables clean data collection. Sequence quality is similar to or better than the manual methods and offers the distinct advantage of a truly hands-off protocol. Details of Beckman Coulter’s Biomek 2000 Laboratory Automation Workstation methods and data generated from these methods are discussed in this poster.
