Ph.D., and Paul Otto The abundant use of PCR products in high-throughput sequencing and microarray research provides the need for automated purification methods. Previous methodologies have employed either centrifugation or vacuum filtration with filter plates to capture, purify and eventually elute PCR products away from excess primers, unincorporated dNTPs and salt contaminants. MagneSil paramagnetic particles have been adapted for purification of amplification products thereby eliminating the need for vacuum collars and filter plates as the process can be performed in standard microplates. The magnetic particles can be manipulated (aspirated and dispensed) on robotic platforms making them particularly suitable for automation. Furthermore, the small particle size ensures rapid sedimentation when placed in a magnetic field.
