The demand for automated methods for plasmid purification has been increasing, therefore necessitating automation in keeping with high throughput assays such as DNA sequencing. To this end, methods have been developed on the Biomek 2000 Laboratory Automation Workstation using Wizard MagneSil paramagnetic particles (PMPs) and reagents available from Beckman Coulter, Inc. Previous methodologies have employed filtration techniques requiring the use of vacuum manifolds and filter plates. However, magnetic particle technology allows for the generation of highly pure plasmids without the need for such specialized labware and filter plates.

Features

  • Utilizes an integrated DPC MicroMix* 5 orbital shaker to efficiently resuspend the paramagnetic particles.
  • Excellent recovery of plasmid DNA, ranging from 6-9 µg.
  • To purify a single plate of plasmids it takes approximately 1 hour and 50 minutes.
