The Sigma Extract-N-Amp Blood PCR procedure utilizing the Biomek 2000 Laboratory Workstation enables users to rapidly extract and amplify human genomic DNA from up to 96 samples of whole blood with minimal user intervention. Briefly, 5µL of whole blood is mixed with 10µL of Extraction Solution, and the mixture is incubated at room temperature for 5 minutes. After adding 90µL of Neutralization Solution, the extract is ready for PCR. An aliquot of the neutralized extract is then combined with the Extract-N-Amp Blood PCR Ready Mix and user-provided PCR primers to amplify target DNA. The PCR master mix comes in two formulations: Extract-N-Amp Blood PCR mix and REDExtract-N-Amp* Blood PCR mix. 

The Extract-N-Amp Blood PCR Ready Mix is a 2X reaction mixture containing buffer, salts, dNTPs, and Taq polymerase. It also contains TaqStart* antibody for hot start PCR to enhance specificity. The REDExtract-N-Amp Blood PCR kit also contains a tracking dye that allows for convenient direct loading of PCR reactions onto agarose gels for analysis.
