Workshop: "From Tissues to Targets: Removing Bottlenecks from the Proteomic Process."

The workshop illustrates how the use of Beckman Coulter’s Biomek® Laboratory Automation Workstations, the Biomek 3000, the Biomek NX and the Biomek FX can remove some of the bottlenecks in the proteomic process. The workshop provides an overview of automation with specific attention to the three workstations from Beckman Coulter. The automation of the proteomic process covered in the workshop focuses on (1) sample preparation, (2) proteome profiling and protein identification, (3) protein characterization and (4) protein functional analysis:

• The methods of sample preparation described include flow cytometric isolation of cells, and subcellular fractionation via centrifugation.

• The use of automation to address the needs of proteome profiling and protein identification are illustrated with a discussion of how the Biomek FX can be used to interface Beckman Coulter’s ProteomeLAB PF 2D with MALDI-TOF MS. The results from mammalian cell culture lysates, solid animal tissues, liquid animal tissues, E. coli lysates, yeast cell lysates and plant tissue lysates are illustrated.

• Protein identification can be facilitated by use of the ProteomeLAB PA 800 for SBS MW analysis and peptide mapping.

• The use of automation for the process of protein characterization is illustrated by the generation of purified proteins for use in an in vitro assay. The process is taken from the preparation of DNA from plasmid expression clones to purification of His-Tag labeled proteins and analysis using an ELISA assay.

• The use of automation for demonstration of protein functional analysis is shown by the preparation of cells and measure of apoptotic response to compounds using the CellProbe HT Caspase 3/7 whole cell assay kit from Beckman Coulter.
