Improving Pipetting Accuracy through Technique Calibration with the Biomek® FX Liquid Handling System

With the increased use of laboratory automation in drug discovery, various ways of determining accuracy of a dispensed volume have been developed. Although gravimetric methods are common, they are not suitable for volumes <10 µL due to volume loss from evaporation. Therefore, a procedure using a spectrophotometer and the calibration feature of the Biomek FX software was developed. 

This procedure allowed modified Scaling Factor and Offset values to be calculated and then entered in the calibration feature of the technique. When the calibration feature was modified using this procedure, the accuracy of dispensed volumes improved. The results presented here demonstrate that laboratories can use a spectrophotometer and the calibration feature of the Biomek FX Liquid Handling System to improve the accuracy of dispense volumes between 0.25 and 1.00 µL.
