The DNA Prep reagent system and workstation offers a simple, semi-automated device for standardized preparation of single cells or nuclei suspensions for the measurement of cellular DNA content by flow cytometry. The operator loads the sample onto the DNA Prep Workstation, closes the protective sample station door and the sample is lysed and stained. It employs a unique lysing and permeabilizing agent which maintains intact cells, adding the dimension of light scatter measurements to traditional DNA analysis. The propidium iodide (PI) stain contains stable RNase to prevent staining of double-stranded RNA. The PI concentration is optimized for the suggested cell concentrations.
