BTS-450 is a quaternary ammonium hydroxide (0.5N) in toluene. It rapidly and economically digests tissues and tissue homogenates. Up to 0.45 mL of water can be dissolved in 1 mL BTS-450. Just follow a simple series of steps to solubilize and count:
- Weigh sample in a glass vial (see table below)
- Add 1 mL BTS-450 and digest sample at 40°C - 50°C until tissue dissolves
- Decolorize with 30% H2O2
- Add 70 µL glacial acetic to eliminate chemiluminescence
- Add 10 mL Ready Organic™ and count
