Agarose slab gel electrophoresis has long been the typical method used
for the analysis of DNA and RNA. Although well established, it is time
consuming, labor intensive and only semi-quantitative at best. Capillary
gel electrophoresis offers an automated approach to nucleic acid
analysis with on-line detection that is fully quantitative, extending
the analytical capabilities of electrophoresis. Gels exist as polymers
in solution, allowing the user an environment which can easily be
optimized. Polymers in solution also allow for an automated mechanism of
gel loading and gel replacement. Beckman Coulter offers the following
nucleic acid kits useful for the routine analysis of oligonucleotides,
viral load determination, gene expression and the quantitative analysis
of DNA-protein interactions through mobility-shift assays.

eCAP ssDNA 100-R Kit
|

|
The eCAP ssDNA Kit is designed for the analysis of oligonucleotides,
providing a direct quantitative tool for quality control. This chemistry
utilizes linear polyacrylamide in solution to form a sieving gel, which
provides single base resolution of oligonucleotides between 10 and 100
bases. The neutral capillary provided with the kit has a stable,
physically bonded polyacrylamide coating, which eliminates
electroosmotic flow (EOF) and suppresses interactions of the sample with
the capillary wall. This chemistry utilizes a standard Tris-Borate EDTA
buffer system including high concentrations of urea to prevent secondary
structure formation. Applications of this kit have included DNA and RNA
probes, ribozymes and anti-sense phosphorothioate oligonucleotides. The
system is compatible with both UV and LIF detection systems to include
the assessment of fluorescent labels.
Kit includes coated capillaries (2), replaceable gel, and standards to
perform at least 70 runs. Urea denaturant is included separately,
enabling a naturing replaceable gel, if so desired.

The eCAP dsDNA 1000 Kit is designed for the analysis of double-stranded
DNA fragments from 72 to 1000 base-pairs. This chemistry utilizes linear
polyacrylamide in solution to form a sieving gel. A neutral capillary
provided with this kit has a stable, physically bonded polyacrylamide
coating which eliminates electroosmotic flow (EOF) and suppresses
interactions of the sample with the capillary wall. This chemistry
utilizes a standard Tris-Borate EDTA buffer system and allows the
addition of intercalating dyes like ethidium bromide or our proprietary
EnhanCE™ to improve resolution and increase sensitivity. This kit is
ideal for the analysis of PCR* products which can be analyzed directly
with no sample clean-up. Applications of this kit have included gene
expression analysis (competitive PCR), direct hybridization viral load
determination (no amplification) and genotyping experiments with RFLPs,
STRs and AmFLPs. The system is compatible with both UV and LIF
detection, but LIF is recommended for most applications.
Kit includes: (2) 100 µm I.D. neutral capillaries (DNA), (3) vials of
replaceable polyacrylamide gel, (1) test mix consisting of Hae III
restriction digest f174 DNA containing 11 fragments from 72 bp to 1,353
bp, and instructions and methods for system set-up and analysis. For LIF
applications, we recommend the use of the intercalating dye EnhanCE.
